Instead, I received another call from them this evening (a different operator), so instead of refusing I simply made up answers to see just what sorts of questions they would ask. It was amazing how blatant they were.&lt;br /&gt;The number shows up on my caller id as 999-999-1234. The caller has a thick accent, and the connection is very 'crackly', like a cell phone in a bad area. They already have my name and address when they call.&lt;br /&gt;They told me they were calling from "Washington Financial Grant Services", and that they have been contacted by the "Federal &amp;amp; State Grant Commission" to distribute grants. I have already been approved for an $8000 grant, simply because I have been paying taxes 'all these years'. The schpiel is that the government puts 10% of all of our taxes into this fund, and the government picks random taxpayers to give 'grants' to out of this fund. Of course, it's a "once in a lifetime" thing and the deadline is "today" (even though they have now called me twice). They even gave me another 'customer service' number that I could call if I had any questions after the call, it was 866-623-4123.&lt;br /&gt;They want to send me 'documents' to claim my grant, but before they can they need to 'verify' that my information is accurate and that I really do have an account. They keep insisting that they'd hate to see me 'lose my money', and keep telling me it happens all the time.&lt;br /&gt;They asked my Date of Birth, How long have I lived at this address, How long have I had this phone number, My bank's name and street address, the bank tracking number, my account number and my mother's maiden name.&lt;br /&gt;I'm afraid it started to fall apart by this time, since I was making up answers (there was no way I'd give them anything even close to the true answers), but apparently they have a way to verify the bank's tracking number and they (of course) couldn't verify the phony number I gave them. "Now it's really a question of marketing and running (the Trump casinos) beautifully."&lt;br /&gt;The company faces stiff headwinds — more than $1.5 billion in debt, better-financed competitors with a big head start on offering the kind of Las Vegas-style resort appeal that Atlantic City has only recently embraced, and at least two deep-pocketed companies who will open new casinos within four years.&lt;br /&gt;It sought buyers for the Taj Mahal, Trump Plaza Hotel and Casino, and Trump Marina Hotel Casino. A proposed deal came close but fell through in July.&lt;br /&gt;Donald Trump said there are still several potential buyers, either for the company as a whole or for a piecemeal sale. But he refuses to identify them or characterize how serious the talks have become.&lt;br /&gt;In the meantime, the company is focused on turning around its bottom line.&lt;br /&gt;"Our turnaround strategy has always been to identify our better customers and market to them," said Mark Juliano, a 30-year veteran casino executive who once ran Caesars Palace in Las Vegas and who took over as Trump's chief executive this summer. "It's to try to get the company away from depending on the day-trip gambler as opposed to someone who wants to come for a two- or three-day entertainment experience."&lt;br /&gt;To do that, Trump needs more hotel rooms to put them in. The company is building a second tower at the Taj Mahal for $255 million that should be open in the third quarter of 2008. The 786 rooms it includes will give Trump 3,670 rooms — about the same as a new casino planned here by Revel Entertainment Group LLC.&lt;br /&gt;The new rooms at the Taj are a major part of the company's strategy to whittle away the $1.5 billion in debt that has limited its options in the past and scared off some potential buyers. The debt remains despite a 2005 bankruptcy reorganization.&lt;br /&gt;Virtually every room in all three casinos has been renovated in recent years, and new restaurants opened at the Taj and Marina. A makeover stripped much of the garishness from the Taj, including its infamous purple carpet, which Juliano promised the state Casino Control Commission "has been banished forever."&lt;br /&gt;Brian Fennell of Lumberton, N.J., stayed at the Taj recently for a trade show, and liked what he saw.&lt;br /&gt;"The decor is fresh," he said. "It looks new and up-to-date. I had a great time."&lt;br /&gt;The company scrapped players-club cards it had issued for each individual casino in favor of the TrumpONE card, which lets gamblers accrue and redeem perks at any of the casinos, even as it cuts back on those for less profitable gamblers. And it hopes to reduce expenses by $19 million this year.&lt;br /&gt;So, will it all work?&lt;br /&gt;Trump Marina and Trump Plaza regularly rank at or near the bottom of Atlantic City's 11 casinos in terms of monthly revenue. And last month, Trump Entertainment reported that its second-quarter loss widened as gambling revenue fell, partly due to increased regional competition.&lt;br /&gt;But the Taj remains around the top third of Atlantic City casinos in monthly revenue and the casino commission recently granted a new five-year operating licenses for the three Trump casinos despite expressing deep skepticism over the company's rosy financial predictions.&lt;br /&gt;Some analysts say that with land and construction prices pushing the cost of new Atlantic City casinos to the $2 billion range, it might be cheaper to buy an existing one rather than build from scratch. Under such a scenario, the Trump properties might look more attractive.&lt;br /&gt;Earlier this month, KeyBanc Capital Markets cited "excellent" progress on reducing promotional expenses such as freebies for gamblers. And Goldman Sachs wrote in an August report that out-of-state competition is hurting the Trump casinos, but noted that the TrumpONE card program and the Taj expansion should increase revenues.&lt;br /&gt;Researchers found that a half-year exposure to meditation techniques helped patients shave as much as one-third of their psychological distress.&lt;br /&gt;The research team, led by Elizabeth K. Pradhan of the University of Maryland School of Medicine's Center for Integrative Medicine, based its conclusions on an analysis of a specific training course called "Mindfulness-Based Stress Reduction" (MBSR).&lt;br /&gt;Pradhan and her colleagues said the MBSR concept of "mindfulness" is designed to guide patients to focus on the "here and now," while emphasizing the value of calmness, clarity, well-being, and a compassion for oneself and others.&lt;br /&gt;The Arthritis Foundation has said that several so-called "mind-body practices" may help arthritis patients alleviate stress, pain, anxiety and depression.&lt;br /&gt;In addition to meditation, the organization acknowledges the potential of electronic monitoring techniques such as biofeedback; the mental health benefits of guided imagery and hypnosis; and the ability of physical activities such as yoga and tai chi to both calm and energize patients.&lt;br /&gt;The Maryland researchers noted that prior studies had already shown that the MBSR course, in particular, seems to have a positive impact on the psychological symptoms of patients with conditions such as anxiety disorders, chronic pain, fibromyalgia, cancer and multiple sclerosis. It was the first-ever analysis of MBSR and its impact on depression, general well-being, and disease progression among rheumatoid arthritis patients, the researchers said.&lt;br /&gt;Thirty-one patients were offered the mediation course over an eight-week period, followed by a four-month maintenance program. At that time it was known as the Tower of Orderly Administration (Qizhenglou).&lt;br /&gt;In 1420, under the Ming Emperor Yongle, the building was reconstructed to the east of the original site and in 1800 under the Qing Emperor Jiaqing, large-scale renovations were carried out. In ancient days, the Drum Tower was the time keeping center for the whole city and was equipped with bronze water clocks and drums that were beaten to mark the hours. The water clocks were later replaced with other timekeeping arrangements. The upper story once housed housed 24 drums, of which only one survives. Its head is made of an entire ox hide and is 1.5 meters in diameter.&lt;br /&gt; A sword slash on the side of the drum was made by soldiers of the Eight-Power Allied Forces during their invasion of Beijing in 1900. In the Qing Dynasty, the hours were marked at night beginning at 7:00 p.m.when the drums were sounded 13 times. After the watch had been "set" in this fashion, each subsequent two-hour interval was marked by a single drum beat. Civil and military officials oriented their lives around these time signals. At the sounding of the third watch (1:00 a.m.) officials attending the morning court audience rose from their beds, and at the fourth (3:00 a.m.) assembled outside the Meridian Gate (Wumen).&lt;br /&gt;At the sounding of the fifth watch (5:00 a.m.) they entered the Imperial Palace and knelt on the Sea of Flagstones (Haimen) before the Hall of Supreme Harmony (Taihedian) to await instructions from the emperor. Close behind the Drum Tower stands the Bell Tower, which first came into use during the reign of the Ming Emperor Yongle. It was destroyed by fire after only a brief existence and it was not until 1747 that Emperor Qianlong undertook its reconstruction. The Bell Tower originally housed a huge iron bell. But because its tolling was not loud enough, this was replaced by a massive cast bronze bell over 10 inches thick that is in perfect condition today. The iron bell was moved to the back of the Drum Tower where it has remained for over 500 years. As recently as 1924, the bronze bell could be heard ringing out the 7:00 p.m. chime from a distance of over 20 kilometers.&lt;br /&gt;According to legend, an official named Deng was given the task of casting the huge bell, and tried unsuccessfully for over a year to carry out the task. As he made one last attempt, his daughter, fearing that her father would be punished if he failed again, decided to sacrifice her life in order to move the gods to bring about a perfect casting, and threw herself into the molten bronze. Her grief-stricken father could only rescue one of her embroidered slippers from the crucible. The casting succeeded and the emperor, moved by the young girl's sacrifice, named her the "Goddess of the Golden Furnace" and built a temple in her honor near the foundry.&lt;br /&gt;After the bell was installed, the chimes could be heard right across the city. But on stormy nights, the bell would emit a desolate moaning sound similar to the word xie, which means "shoe" in Chinese. Leona sang her heart out on top of aladder, on top of a bus whilst 20 contemporary dancers did theirstuff way below. It was just so low key relative to what had goneon beforeit.　&lt;br /&gt;　But oh, our offering at the closing ceremony was embarrassing.Ared bus,a pop singer, an ageing rocker and an overpaid ball kickerwas awful. (Clip off handle of brush with wire cutters so tube can be closed. Cutters should be rinsed with EtOH between samples to prevent contamination.)2. Vortex thoroughly to mix, for at least 10 s.3. Heat to 95∫C for 5 min.4. Spin briefly to pool condensation.5. Remove brush with tweezers (tweezers should be rinsed with ethanol between samples to prevent contamination) and add 60 ml 1M Tris pH 8.0 to tube. Vortex to mix for at least 10 s.6. Spin for 1 minute at 13,000 rpm and decant supernatant to a clean storage tube.7. To celebrate, we looks back the key scientific achievements of the twentieth century, forward to DNA's future, and around at the double helix's place in biology.Protocol for extracting DNA from ES Cells, starting from the 96-well plate but processing in an eppendorf tube to recover more of the DNA. NOTE- THIS TAKES A LOT OF TIME if you do the whole plate this way!Lysis buffer: ( 100 ml Recipe )10 mM Tris-HCl pH7.5 - 0.5 ml of 2M10 mM EDTA - 2 ml of 0.5M10 mM NaCl - 0.2 ml of 5M0.5% (w/v) Sarkosyl - 0.5gm (N-Lauroylsarcosine, Sigma # L-9150)1mg/ml Proteinase K - add fresh each time (stored in freezer).1. Add 250µl Lysis Buffer (with proteinase K added) to each well.2. Incubate the plate in a sealed humid container at 60°ree;C for 1 hour. (Meanwhile, label eppy tubes!)3. Transfer the contents of each well into a separate 1.5ml eppendorf tube (µ-cent. tube).4. Continue lysing the cells for 2 more hours. (60°ree;C). (Float tubes in waterbath- don't need tupperware).5. Add an equal volume (250 µl) of phenol:chloroform (phenol: chloroform: isoamylalcohol, 25:24:1).(The pH of the phenol:chloroform needs to be ~7.8 - 8.0, or else the DNA will partition into the organic phase).6. Mix the contents of the tube until an emulsion forms. (Shake by hand for ~1 min, (or vortex ~2sec),because vortexing shears long (genomic) DNA).7. Centrifuge 5-10 min in a microfuge at TRm. (The phases should be well-separated).8. Transfer (pipet) the aqueous phase (the top phase) to a fresh tube. (Toss the interface/organic phase-- chemical waste in hood).9. Repeat steps 5-8 until no protein is visible at the interface of the aqueous and organic phases.10. Add an equal volume (250 µl) of chloroform and repeat steps 6-8.11. Optional: Repeat chloroform extraction. (The chloroform extraction removes the traces of phenol).12. Add 1/25th volume of 5M NaCl (10µl of 5M NaCl for 250 µl ), for a final concentration of 0.2M NaCl. Mix well. (use NaCl rather than other salts because of detergent in lysis buffer).13. Add exactly 2 volumes of ice-cold ethanol and again mix the solution well.14. Store the ethanolic solution on ice for 30 min to precipitate the DNA. (can be stored indefinitely at 0°ree;C or at -20°ree;C).15. Centrifuge at 12,000xg for 10 min, 0°ree;C. (Try 4°ree;C, microfuge at max (16,000), 10 min). (Remember to point tube hinges out, to locate pellet even if it's invisible).16. Suck off the supernatant (dispo pipet tip, with vacuum flask). Do not disturb the pellet. Vacuum droplets from walls of tube as well.17. Half fill the tube with 70% EtOH and re-spin for 2 minutes at 4°ree;C.18. Suck off supernatant as in step 16.19. Store the open tube on the bench at TRm until the last traces of fluid have evaporated. (i.e.: Air dry).20. Dissolve the DNA in 30µl (or 50 µl, and use half of sample) of 10mM Tris pH 8.5 (or TE). (pipet around the walls of the tube to dissolve the DNA off the walls of the tube). Let dissolve for at least an hour at 37°ree;C.21. Restriction digest:- DNA in Tris pH8.5, from above protocol- restriction enzyme- restriction enzyme buffer- X (BSA if required by enzyme)- spermidine (to a final conc. of 1mM; stored in freezer)- (also add: ) DTT to a final conc. of 1mM Final volume = 40µl or less. Digest overnight at 37°ree;C.Next: Run gels; Southern blot and hybridize. Today, he is far from sure that's still the case. Along with an increasing number of other well educated Chinese living in America, Lee can't wait to return home as China, with its turbo-charged economic growth, offers better job prospects than the United States. Even the outbreak of the deadly SARS virus in Hong Kong and the mainland does not appear to be a deterrent.Lee said he quit his job to earn an MBA at MIT to help him get on the corporate fast-track back home. He envies his former classmates from a top university in China, as they now hold top corporate positions.To be sure, there are still more Chinese coming into the United States than are returning, but a recent survey found that 80 percent of Chinese students now studying abroad said they would like to go back.Overall, China still suffers more of a brain drain than a brain gain: In the last 25 years, only about a quarter of the students have returned home after graduation, according to the Chinese Ministry of Education.But the number of "sea turtles," the nickname for returnees to China, is growing rapidly. An example was former GOP Majority Leader Dick Armey, saying last week that "James Dobson and his gang of thugs are real nasty bullies," labeling the Focus on the Family leader as engaging in "high demagoguery." Since Armey is now President of FreedomWorks, one of the key corporate-backed lobbying groups in DC and in statehouses around the country, this is a serious declaration of war between previous political allies.The Evangelical Conflict over Climate Change:&lt;br /&gt;But the most serious conflicts are now happening within the religiously conservative movement itself. A clear example was when evangelical leaders this year created the Evangelical Climate Initiative to address climate change and clean up our environment, based around a statement signed by 86 initial evangelical leaders, including Rev. Dr. Leith Anderson, Former President, National Association of Evangelicals (NAE), Rev. Ron Sider, President, Evangelicals for Social Action and Rick Warren, pastor of Saddleback Church and author of mega-seller The Purpose Driven Life.&lt;br /&gt;This was organized by the even older Evangelical Environmental Network, which includes a range of groups including the Texas Baptist Convention.Denouncing this effort in an open letter were other rightwing evangelicals tied to the more traditional political rightwing, including James Dobson, Richard Land of the Southern Baptist Convention, former Watergate criminal, now religious leader Charles Colson, Lou Sheldo of the Traditional Values Coalition, and Donald Wildmon of the American Family Association.Other Divisions Among the Religious Right: This conflict forced the National Association of Evangelicals (NAE), which represents 51 denominations and 10 million people, to abstain from taking a stand on global warming. In a sign of other divisions, the NAE took no position when the war in Iraq started due to the stalemate between anti-war and pro-war factions.Similarly, when Dobson and others on the far right objected to US funding for the Global Fund to Fight AIDS, Tuberculosis and Malaria, because a small portion of its funds go to distributing condoms and providing clean needles to drug addicts, evangelical leaders like Rick Warren and even Pat Robertson stood up to Dobson in defense of the Global Fund as critical for saving lives around the world.The "Other" Baptists: As the Southern Baptist Convention moved sharply to the theological and political right in the last few decades, dissent has broken out throughout its institutions. In 1991, a large number of Southern Baptists formed the Cooperative Baptist Fellowship, including 1800 partner churches and 13 theology schools and seminaries, as an alternative to the rigid fundamentalism of the SBC.&lt;br /&gt;An even more progressive group of Southern Baptists have broken off to form Mainstream Baptists.Most significantly, leaders of these dissident Southern Baptist groups have begun a dialogue with the northern American Baptists (from which the Southern Baptists split in the 19th century over the issue of slavery) and with the African-American Baptist denominations. This year, former President Jimmy Carter (a former Southern Baptist himself) convened a meeting where various Baptist leaders committed to exploring building a North American Baptist Covenant dedicated to "promote peace with justice, to feed the hungry, clothe the naked, shelter the homeless, care for the sick and the marginalized, welcome the strangers among us, and promote religious liberty and respect for religious diversity." John 1:1-3, Eph 3:9, Col. 1:12-16. God rested because His work was finished. That is the only justifiable reason for resting.&lt;br /&gt;God rested because His work was not only finished but was GOOD. There could be no reflection upon it. But when God’s perfect work was marred by sin, by the “Fall of Man,” His “Sabbath Rest” was broken. As soon as man fell it was necessary for God to resume His work, this time not to continue the creation of material things, but for the purpose of the redemption of man that he might become a “NEW CREATION” in Christ Jesus. 2 Cor. 5:17. So Jesus, in explaining His mission said –"My Father is working until now [in creation], and I Myself am working [now in REDEMPTION]." John 5:17.In Gen. 2:1-3, we are simply told that God rested from His work of creating on the ‘SEVENTH Day.” The Day is not called a SABBATH Day.&lt;br /&gt;We are told that God BLESSED and SANCTIFIED it. That is, set it apart as a REST DAY. We are not told the length of the “SEVENTH Day” of the Creative Week, but it should have corresponded with the length of the other days, if sin had not shortened it.THE SABBATHIf God instituted the “Sabbath” before the “Fall of Man,” it seems strange that the fact is not recorded in Genesis, and that Adam was not told to observe it. Nowhere in the Book of Genesis do we read of Adam, or any of his descendants, or Noah, or Abraham observing the Sabbath. The only hint we have of a “seven-fold” division of days is found in Gen. 7:4, 10, when seven days of grace were granted before the Flood came, and in Gen. 8:8-12, where a seven-day period elapsed between the sending forth of the dove.&lt;br /&gt; The first place we read of the Sabbath is in Ex. 16:23-26, in connection with the gathering of the manna -- Six days you shall gather it, but on the seventh day, the Sabbath, there will be none." Here we have the “SEVENTH” day designated as “THE SABBATH.” That the “Seventh Day” of the “Creative Week” was a type of the Sabbath is clear from Ex. 20:11 – “For in six days the LORD made the heavens and the earth, the sea and all that is in them, and rested on the seventh day; therefore the LORD blessed the Sabbath day and made it holy.” But we have no evidence that the Sabbath was commanded to be observed until after the Exodus, and the reason is clear. God’s “Rest Day” was broken by the “Fall of Man,” and there could be no “rest” until redemption was brought in, and this was typically brought in by the redemption of the Children of Israel from Egypt through the offering of the “Passover Lamb,” a type of Christ. The purpose of their deliverance was that they might find rest in Canaan from the weary toil and slavery of Egypt. Deu. 5:15.When a few weeks later the “Ten Commandments” were given on Mt. Sinai the Lord said to Israel, “Remember the Sabbath day, to keep it holy,” (Ex. 20:8), the Sabbath Day they were to remember was not the “Seventh Day” on which God rested, but the “Day” that God had appointed as the “Sabbath Day” at the time of the giving of the manna.The command to observe the Sabbath was given to Israel EXCLUSIVELY. It was not given to the Gentiles. It was given to Israel as the “SIGN” of the “Mosaic Covenant.” “But as for you, speak to the sons of Israel, saying, 'You shall surely observe My Sabbaths; for this is a sign between Me and you throughout your generations.” Ex. 31:13; Ezek. 20:12, 19-21.&lt;br /&gt;The Sabbath Day then belongs to the Jews alone and is not binding on the Gentiles (the world), or on the Church (Christians). Nowhere in the Bible do you find God finding fault with any nation or people, except the Jewish nation, for not observing the Sabbath. As a Jewish ordinance it has never been abrogated, changed, or transferred to any other day of the week, or to any other people. It is now in abeyance as foretold in Hosea 2:11 it would be. It is to be resumed when the Jews are nationally restored to their own land. Isa. 66:23. Ezek. 44:24; 46:1-3.If this be true, then the “Sabbath’ does not belong to the Church, and is not to be observed by Christians, for the “Sabbath Day” is a part of “THE LAW,” and Christians are nut under “LAW,” but under “GRACE.” Rom 6:14. In his letter to the Galatian Christians Paul reproved them for going back to the “Law,” and declared that those who did so were “under the CURSE.” Gal 3:10. “But now that you have come to know God, or rather to be known by God, how is it that you turn back again to the weak and worthless elemental things, to which you desire to be enslaved all over again? You observe days [Sabbath and Feast Days] and months and seasons and years. I fear for you, that perhaps I have labored over you in vain.” Gal. 4:9-11. “Therefore no one is to act as your judge in regard to food or drink or in respect to a festival or a new moon or a Sabbath day—Col. 2:16. If Christians are under obligation to keep the “Jewish Sabbath,” then they are under the Jewish “Ceremonial Law” and should observe all the ordinances and Feast Days of the Jewish Ritual.As an institution of Judaism, the Sabbath, with all the “Feast Days” and other ritualistic ceremonies and offerings of Judaism, ceased to function with the close of the Jewish Dispensation. The JEWISH Sabbath was not changed to the CHRISTIAN Sabbath, any more than “Circumcision” was changed to “Baptism.” There is no such thing as the “CHRISTIAN Sabbath.” “Sabbath” has to do with LAW, and “Christian” with GRACE, and to join “LAW” and “GRACE” is to unite what God has forever separated.After the Resurrection, Christ and His Disciples never met on the “Sabbath” but on the “FIRST DAY of the week.” John 20:1, 19. Acts 20:7. 1 Cor. 16:2. It is true that they went into the Jewish Synagogues on the Sabbath, but not to worship, but that they might have opportunity to preach the Gospel. The “First Day of the Week” is the day to be observed for rest and worship by the Christian Church. It is prefigured in the Old Testament as the “EIGHTH DAY,” or the “DAY AFTER THE SABBATH.” “You shall bring in the sheaf of the first fruits of your harvest to the priest. 'He shall wave the sheaf before the LORD for you to be accepted; on the day after the Sabbath the priest shall wave it.’” Lev. 23:10-11.&lt;br /&gt; What did that “First Fruits” typify? Read 1 Cor. 15:20 – “But now Christ has been raised from the dead, the first fruits of those who are asleep.” When did Christ rise from the dead and become the “FIRST FRUITS?” Not on the “Sabbath,” for He lay dead in the tomb on that day, but on the “FIRST DAY OF THE WEEK,” on the “DAY AFTER THE SABBATH.”The fact that the “Birthday” of the Church was at Pentecost, and that fell on the “First Day of the Week,” is further proof that the Church should keep the “First Day of the Week” and not the “Seventh” day or “SABBATH.”The Jewish Sabbath links man with the “Old Dispensation,” the “First Day of the Week” links man with the “New.” It is claimed that the Roman Catholic Church changed the day of rest from the “Seventh Day” to the “First Day of the Week,” but the claim is false, for the Papacy did not exist until a long time after the “First Day of the Week” had become a fixed day for Christian worship. It is a noteworthy fact that the whole of the “Ten Commandments” (Ex. 20:1-17) are reaffirmed in the New Testament, except the “Fourth Commandment” regarding the Sabbath. Rom. 13:8-10, Eph. 6:1-2, James 5:12, 1 John 5:21. Why this omission if the Law of the “Sabbath” is still in force?&lt;br /&gt; It is called the “LORD’S DAY.” It belongs to Him. It is not called a “rest day” in the Bible. It is a day that should be filled with worship and service and holy activity.
